CONSTITUTIVE EXPRESSION OF ZINC FINGER TRANSCRIPTIONAL REGULATORY FACTORS BY HUMAN SOLID TUMOR CELL LINES (MEETING ABSTRACT)

Abstract:
Zinc finger genes 225 (EGR-1) and 592 (EGR-2) are rapidly and transiently expressed in cells transiting from G0 to G1. In contrast, T cells transformed by HTLV-I or HTLV-II constitutively express these genes suggesting that dysregulation of these factors plays a critical role in transformation. We investigated solid tumor cell lines, including 4 rhabdomyosarcomas, 5 colon cancer and 12 small cell (SCLC) and non-small cell cancers (NSCLC) of the lung, for constitutive expression of 225 and 592. All rhabdomyosarcoma cell lines and 3 colon cancer cell lines constitutively expressed 225. Approx 25% and 50% of SCLC and NSCLC cell lines, respectively, expressed 225 constitutively. All extrapulmonary SCLC tumors were positive. No correlation between SCLC morphology and expression was noted. Coexpression of 225 and 592 was seen in one NSCLC cell line but no other cells expressed 592. These studies support a role for 225 in malignant transformation of solid tumors, and suggest that alternative pathways for constitutive activation of these genes exist in the absence of retrovirus transformation.
