Abstract: FSA tumor cells that had the mIL7 gene inserted by retrovirus infection (FSA-JmIL7) had decreased tumorigenicity and a high incidence of spontaneous regression. This was associated with a heavy infiltrate of T cells. The number of CD8+ cells was particularly increased, leading to a shift in the balance of CD4:CD8 cells from 1:0.8 in FSA-Jneo to 1:1.6 in FSA-JmIL7. TIL from IL7-producing tumors had greatly enhanced cytotoxicity towards uninfected, parental FSA cells. Killing of non-cross-reacting cells was also increased but to a much lesser extent. There was also an eosinophil/basophil (9-22%) in FSA-JmIL7 that was not present in FSA-Jneo. There were no significant changes in the number of macrophages (10-20%) or B cells (5-8%) as a percentage of total cells, but the ratios of macrophages and B cells to tumor cells were inevitably increased. The number of tumor cells in FSA-JmIL7 varied from 7-33%, presumably reflecting the extent of tumor regression. These findings suggest that IL7 may prove to be a useful cytokine for gene-mediated immunotherapy.
