Abstract: The synthesis and biological evaluation of analogs of two antimitotic natural products were undertaken. Application of a novel manganese(III) acetate oxidation process provided the key step in the synthesis of podophyllotoxin congeners. The syntheses of several A and AB-ring taxol analogs were also accomplished. A tubulin turbidimetric assay was used to evaluate the bioactivity of the synthetic podophyllotoxin and taxol analogs. The brine shrimp toxicity test, platelet-activating factor binding assay, anti-Rhino and anti-Herpes viral assays of the podophyllotoxin intermediates are also summarized. The National Cancer Institute's anticancer and anti-HIV screening results of the podophyllotoxin intermediates are included. X-ray structures and 2-D NMR spectra of pertinent intermediates and products are reported. (Full text available from University Microfilms International, Ann Arbor, MI, as Order No. AAD91-27664).
