DETECTION OF NOVEL ETIOLOGICAL AGENTS AND HERPESVIRUSES (MEETING ABSTRACT)

Abstract:
Human herpesvirus-6 (HHV-6), human herpesvirus-7 (HHV-7) and B-lymphoma-associated virus (BLAV) are lymphotropic herpesviruses, which were discovered in the last few years. HHV-6, discovered in 1986 by Salahuddin et al, has been classified as a lymphotropic herpesvirus. The virus readily infects mitogen activated cord and peripheral blood cells in vitro. Certain T-cell lines can also be infected with the virus, however, nearly every strain of HHV-6 isolated so far requires a distinct type of T-cell line for replication. It has also been reported that HHV-6 may transform 3T3 fibroblast indicating that the virus may have oncogenic properties. In our laboratory, we successfully established several cell lines containing the latent HHV-6 genome by infection of established T-cell lines, peripheral and cord blood cells, in vitro with HHV-6. These cell lines express new T-cell markers on their surface which are not present on normal T-lymphocytes. HHV-7, another lymphotropic herpesvirus, was discovered in 1990 by Frenkel et al. This virus shares homologous DNA sequences with HHV-6 and the characterization of the virus is under progress. The newest human herpesvirus, BLAV, has been isolated from an EBV genome-negative Burkitt's lymphoma cell line. The virus is distinct from any known human herpesviruses based on DNA hybridization data and serological peripheral blood B cell in vitro. The possible role of HHV-6 and BLAV in AIDS and various lymphomas is discussed.
