Abstract: The CD4 molecule is a single-chain transmembrane glycoprotein of 55 kD expressed on a broad range of cell populations, including thymocytes, peripheral T subsets (helper/inducer T cells), monocytes, histiocytes and microglial cells. CD4 is the receptor for class II molecules and HIV; its cytoplasmic domain is associated with a protein-tyrosine kinase p56lck; on CD4+ lymphocytes it regulates the function of CD3/TCR complex. The wide range of cellular expression of CD4 molecule prompted us to investigate its possible presence in blast cells in acute myeloid leukemia. We studied 15 AML cases by cytofluorimetric analysis with MoAb Leu3 PE and by total cellular RNA extraction, Northern blot and hybridization with a specific probe. We found that the majority of investigated cases (12/15) expressed full-length CD4 mRNA according to immunophenotyping data presented in a table. Thus, AML blast cells we studied can transcribe, translate and express on their cell surface the CD4 molecule, which might be considered to belong to their phenotypic repertoire.
