Abstract: It has been previously shown that recombinant Tat protein (rTat), a HIV gene product that is necessary for viral replication in vitro, inhibits antigen- but not mitogen-induced lymphocyte proliferation. We have evaluated the immunosuppressive effects of rTat protein on macrophage (Mphi)-mediated tumor cell killing. Addition of rTat protein to the murine peritoneal Mphi cell line DJ2PM-PC results in a dose-dependent inhibition of tumoricidal activity induced by the combination of IFN-gamma and LPS. Furthermore, homotypic aggregation of the cells becomes evident after treatment of the Mphis with rTat protein and activators. Polyclonal antibody against rTat protein, but not a control serum, reversed both the inhibition of Mphi cytotoxicity and the cell aggregation, demonstrating that these phenomena are specifically due to the rTat protein. These novel properties of rTat protein may contribute to immunosuppression and pathogenicity in HIV infection.
