REGULATION OF NF-KAPPA B/REL ACTIVITY IN T CELLS EXPRESSING THE TAX ONCOGENE OF HTLV-I (MEETING ABSTRACT)

Abstract:
The NF-kappa B/rel transcription complex represents a family of DNA-binding proto-oncogenes involved in transcriptional regulation of many immunomodulatory cytokines and cell surface receptors. The effect of the HTLV-I tax oncogene on NF-kappa B DNA-binding activity and NF-kappa B-dependent transcription was examined in tax-expressing Jurkat T cells. Stable expression of tax resulted in constitutive transcription of the endogenous GM-CSF gene and increased basal level of expression of transfected NF-kappa B-regulated promoters. In situ UV-cross-linking analyses of NF-kappa B protein-DNA complexes revealed the presence of several polypeptides ranging from 58 to 92 kD. Multiple forms of NF-kappa B were purified by DNA-affinity chromatography and shown to possess distinct DNA binding and gene activation properties using an in vitro transcription system. We are now investigating the interactions of recombinant NF-kappa B p50, p65 and c-rel with the tax protein and with the regulatory subunit I kappa B (inhibitor of NF-kappa B). These studies indicate that tax expression may alter the stoichiometry of NF-kappa B/rel proteins and change the expression of growth-regulated genes.
