Abstract: We isolated a cell line from the pleural exudate of a pt with myeloplastic syndrome exhibiting chronic myelomonocytic leukemia. After 3 generations, in vitro cells were mostly immature T cells (TCR gamma/delta+) containing myeloid marker CD15 but neither CD13 nor CD33. The cells are tumorigenic in nude mice, causing sc lymphoma indistinguishable from cutaneous tumors in the pt. A cell line established from the nude mouse tumor is identical to the original isolate: trisomy 21, CD4-, CD8-, class II Ag-, class I Ag+, CD15+, deoxynucleotide transferase (TdT+), transferrin receptor (CD71+) and progenitor antigens (CD34+). Rag 1 mRNA was detected in the cells. Immature T-cell markers (CD3-, TCR gamma/delta+) were expressed in 63%, and mature T-cell markers (CD3+, TCR alpha, beta+) were found in 10%. The addition of TPA downregulates PKC; cells differentiating in the T-cell lineage were 53% mature (CD3+, TCR alpha/beta+) and 34% immature (CD3-, TCR gamma/delta+). The combination of TPA and Br-cAMP treatment increases the proportion of cells with mature T-cell markers to 90%, all expressing IL-2 receptors (CD25). Transfection with HIV-1 genome cells produces infectious HIV-1 without cell lysis.
