Abstract: OBJECTIVES: To evaluate immune responses to a trivalent recombinant vaccinia virus vaccine which expresses three HIV-1 (IIIB) genes (env, gag, and pol). In addition we evaluated immune responses to HIV-1 pseudovirion particles produced in mammalian cells infected with the trivalent vaccine. METHODS: Rabbits were primed and boosted with combinations of the above described immunogens. Serum neutralizing assays were done against IIIB RF, and MN strains of HIV-1 as well as patient isolates. Neutralization was defined as 90% inhibition of p24 production. RESULTS: All rabbits developed strong antibody responses to core proteins as evidenced by Western blot analysis. Responses to env glycoproteins were weaker. Rabbits given both immunogens produced antibodies which neutralized the infectivity of HIV-1 (IIIB) in vitro. Neutralizing responses to several heterologous strains and patient isolates of HIV-1 were seen in rabbits immunized with the trivalent recombinant vaccinia virus and boosted with pseudovirion particles. A cellular system for evaluation of cytotoxic lymphocyte (CTL) responses was developed using homologous skin fibroblast cell lines as targets for CTL assays. CONCLUSIONS: A recombinant vaccinia virus vaccine expressing multiple HIV-1 genes along with pseudovirion particles derived from infected cells were used together to induce broad spectrum neutralizing antibodies. This trivalent vaccine is a candidate for a phase 1 trial in humans.
