Abstract: Murine AIDS (MAIDS) shows many similarities with human AIDS. The etiologic agent of MAIDS is a defective retrovirus encoding a precursor gag protein (Pr60gag) which remains uncleaved. Helper-free stocks of this defective virus have been shown to induce disease. Using such helper-free stocks, we have found that the infected target cells proliferated markedly following infection, first as a polyclonal expansion which eventually reaches a stage of clonality. Therefore, this disease appears to be primarily a lymphoproliferative disorder. In diseased mice inoculated with helper-free stocks of tagged defective viruses harboring unique sequences, we have identified the target cells of the defective virus by in situ hybridization/immunocytochemistry techniques. Infection of this target cell population appears necessary and sufficient to induce disease. Our data are discussed in the context of immunodeficiency associated with lymphoproliferative disorders (as a paraneoplastic syndrome), and in the context of superantigens, since Pr60gag appears to exhibit some of the characteristics of a superantigen.
