GEOGRAPHY MORE THAN PATHOLOGY INFLUENCES THE LIMITED HTLV-I GENOMIC VARIABILITY (MEETING ABSTRACT)

Abstract:
In order to study the geographic and/or disease-associated variations of the HTLV-I genome, the ex-vivo DNA from seropositive peripheral blood lymphocytes of 4 Japanese patients (pts; 2 with tropical spastic paraparesis/HTLV-1 associated myelopathy [TSP/HAM] and 2 with adult T-cell leukemia [ATL]), 3 Caribbean pts (2 with TSP, 1 with ATL), and 2 Ivory Coast African pts (1 with TSP, 1 with B lymphoma) were studied by PCR, followed by direct DNA sequencing. For each preparation, about 2000 bp were amplified and sequenced, including parts of the LTR (nt 31 to 768) of the env gene (nt 5684 to 6151), and the px II, III and IV coding frames of the px region (nt 7201 to 8440). Minor genomic variations were observed mainly in the U3 sequence of the LTR region and in the px IV coding frame, where numerous single-point mutations were detected. No specific mutation could be linked to either ATL or TSP/HAM. To further assess the existence of possible geographic, LTR-related variants, 30 further HTLV-I seropositive blood lymphocyte DNA samples (17 from Japan, 6 from Caribbean basin, 5 from Africa, 1 from China and 1 from USA) were amplified in their LTR regions (nt 31 to 768) and digested by 6 enzymes having restriction sites within the U3 region. The restriction patterns observed in this series suggest the existence of 2 HTLV-I subpopulations in Japan (one genuine and one of the 'Caribbean' type), and possibly a west-African subtype.
