Abstract: Mycoplasmas of the M. fermentans species, a rarely identified urogenital mycoplasma was shown to infect the kidneys of HIV positive patients with AIDS-associated nephropathy. We set out in this study to 1) detect M. fermentans in the urine sediments of HIV positive patients utilizing the polymerase chain reaction (PCR) and 2) to isolate M. fermentans from these urines. Using the synthetic oligonucleotide primer pairs, RW004 and RW005 which contain specific sequences within the insertion sequence-like genetic element found only in mycoplasmas of the M. fermentans species, 10 of 43 urine sediments obtained from 40 HIV positive patients at different stages of AIDS disease tested positive. M. fermentans was isolated and grown in mycoplasma cultures from three urine sediments obtained from two HIV positive individuals. Fifty urine sediments from age-matched HIV negative healthy controls tested negative for M. fermentans by PCR. M. fermentans was not isolated from any of the control urine samples. Our results show a high prevalence of the AIDS-associated mycoplasma, M. fermentans, in the urine from HIV positive patients but not from HIV negative non-AIDS controls.
