Abstract: PCR is an effective diagnostic tool for the identification of HIV-1 infection from clinical specimens. Due to the presence of cross-seroreactivity and mixed infections in regions of West Africa, the use of PCR to differentiate HIV-1 and -2 infections is important. However, difficulty is encountered in designing HIV-2 specific primers because HIV-2 exhibits a higher degree of genetic variation than HIV-1. Two HIV-2 isolates derived from the Cote d'Ivoire (CI) and the Cape Verde Islands were used for amplification. Lymphocytes from patients residing in CI were tested with primers derived from gag and env of HIV-1 and from the LTR, gag, pol, env and X of HIV-2. Amplified products were detected by hybridization protection assays with chemiluminescent probes or by Southern blotting with 32P-labeled oligonucleotide probes. Most of the HIV-2-specific primer pairs detected many but not all specimens from patients serologically positive for HIV-2. To maximize detection of HIV-1 and -2 infection, the use of multiple primer pairs is necessary.
