Amplification of HIV gag and env genes from kidney biopsy archival material.

Abstract: Patients infected with the HIV virus may develop a form of renal disease known as HIV-associated nephropathy. To elucidate whether HIV could cause renal disease directly, we first examined renal tissue from 8 patients with kidney disease to determine if HIV genome was present. DNA from paraffin sections of renal biopsy was extracted by a modified Higuchi method. A sensitive nested polymerase chain reaction amplified target sequences within the gag and env regions of the HIV proviral genome. Target sequences were detected with 32P end labeled probes. Six of the patients were HIV positive. Four had focal glomerulosclerosis (FSGS), one IgA nephropathy and one had minimal change disease. The two control patients had FSGS but were HIV negative. gag and env target sequences were detected in all six HIV positive patients, but not from the two control patients. HIV genome was found in HIV infected patients with HIV nephropathy. The pathogenic role of viral infection is unknown. Currently, efforts to examine microdissected individual cells for the presence of HIV are in progress.
