Abstract: HIV is the etiologic agent of AIDS, and the demonstration of HIV antigen in patient serum or plasma samples correlates with disease progression. Further, monitoring HIV antigen levels during therapeutic intervention correlates with the therapeutic's efficacy. Thus, a method for the rapid and accurate quantitation of HIV antigen in serum or plasma will be useful as both diagnostic and prognostic indicators of HIV infection. An enzyme immunoassay (EIA) to detect and quantitate Human Immunodeficiency Virus (HIV) in plasma or serum has been developed for use on an automated instrument. The EIA employs paramagnetic microparticles and an enzyme conjugate substrate system. Anti-HIV immunoglobulins, which are attached to the microparticles, are incubated with sample. Following a programmed wash sequence, the conjugated is added and fluorogenic product is quantitated by kinetic measurements.
