Comparison of mucosal vs. intravenous infectious dose of a cell associated SIV32H challenge stock in rhesus macaques.

Abstract:
A rhesus macaque infected with 10 monkey infectious doses (MID50) of the 11-88 stock of SIV32H served as a single source of peripheral blood mononuclear cells (PBMCs) and plasma taken at a single time point to establish a cell associated virus stock for homologous or heterologous vaccine challenge. At the time of collection the monkey showed early signs of disease, a mild plasma antigenemia and a decrease in CD4+/CD29- lymphocytes. PBMCs were found to be more than 90% viable (5 x 10(5) cells/ampule), following freezing and were analysed by FACScan for specific subset. Animals were infected either intravenously (i.v.) or via rectal mucosa with decreasing serial five-fold dilutions beginning at 1 x 10(6) lymphocytes per macaque. Mucosal challenge was controlled by administration of a previously determined cell-free dose of the 11-88 SIV32H. An MID was established by an endpoint dilution as determined by animals which remained sero-negative for SIV as determined by ELISA, virus isolation and PCR three months post-infection. The characteristics of this challenge stock and the minimum infectious dose via mucosal or i.v. route will be presented.
