Abstract: To help meet the demands of in vitro antitumor drug screening, cell lines were established from patient samples or xenograft tumors. Tissue samples were implanted in nude mice, carried for 1-2 passages, and then used in explant primary culture on plastic and extracellular matrices in several media. Growth kinetics, plating efficiency, cellular morphology, HIV, HHV2, MAP, and mycoplasma expression were measured. The lines were adapted to grow in RPMI 1640 media with 10% FBS. Tumorigenicity studies were conducted in nude mice from lines passaged over 5 times. All lines developed were tumorigenic in nude mice. The lines established to date include 1 gastric, 1 colon, 5 lung, and 4 melanoma. Stocks of cryopreserved cells were prepared for possible use in the NCI in vitro antitumor drug screen.
