Abstract: CD4 is a glycoprotein identified as the host cell membrane receptor for the HIV envelope protein gp120. Others as well as we have observed the presence of CD4 on the HT29 colonic cancer cell line. We have observed that the presence of CD4 may be cell-cycle dependent. Demonstration of CD4 in normal and neoplastic intestinal tissues has not been reported. In our work we demonstrate, by immunoperoxidase technique on paraffin sections, the presence of CD4 on normal and neoplastic human colon. CD4 was present prominently in the differentiated surface epithelium and apical portion of normal epithelial cells. CD4 was present on the membrane, as demonstrated by CD4 immunofluorescence, of live cells. This marker was not present universally on all cells, suggesting epithelial cell subpopulations. CD4 was absent in undifferentiated colonic carcinomas, and weakly present on well-differentiated carcinomas. The endothelium of vessels supplying both normal and carcinomatous areas expressed CD4. The demonstration of CD4 on normal intestinal epithelium may help explain infection by HIV in normal epithelium. Decreased presence of CD4 in carcinomas suggests that expression of CD4 is dependent on cell differentiation.
