CLONING AND SEQUENCE ANALYSIS OF THE ENVELOPE GENE FROM THE FV-2(RR)-ADAPTED BSB STRAIN OF FRIEND SPLEEN FOCUS-FORMING VIRUS

Abstract:
Friend spleen focus-forming virus (SFFV) is a replication-defective retrovirus that causes an acute erythroleukemia in susceptible mice. It has been shown that the primary translation product of the SFFV env gene (gp52) is the oncoprotein responsible for induction of the disease. Susceptibility of mice to F-SFFV is controlled by the Fv-2 gene. This gene inhibits both SFFV replication and spleen foci formation. Mice homozygous for the Fv-2r allele (eg, C57BL/6) are resistant to wild type SFFV-induced disease. BSB is an Fv-2rr adapted variant that causes erythroleukemia in resistant mice. The envelope protein of SFFV(BSB) has an apparent Mr of 43,000 (gp43). The env gene of SFFV(BSB) was amplified by the polymerase chain reaction, cloned and sequenced. Analysis of the nucleotide sequence indicated a large 159-base-pair (bp) deletion in the ecotropic-related region of the SFFV(BSB) env gene. The deletion leads to the loss of about two-thirds of the p15E-related domain. Direct linking between non-ecotropic domain (on the cell surface) and the 22-hydrophobic-amino acid stretch (embedded in the plasma membrane) is a unique feature of the SFFV(BSB) gp43 molecule. In addition to the deletion, five amino acid substitutions occurred in the highly conserved domain (amino acid residues 92 to 158) of wild type SFFV envelope proteins.
