Abstract: HTLV-I-producing human lymphoid cell lines, derived from ATL and HAM patients, and lymphocytes from HTLV-I carriers and seronegative healthy individuals were tested for lymphocyte markers, on the viral envelope and the plasma membrane. Short-term cultured lymphocytes from HTLV-I carriers, but not the virus were labeled with CD4. In the MT-2 cell line, still retaining the lymphocyte-transforming ability, both HTLV-I virions and plasma membranes were strongly positive for OKIa1 or HLA-D and CD25, but not for CD3, 5, WT-31 and beta F1. Beta F1 gave positive immunoreactivity with the perinuclear space and rough endoplasmic reticulum in some cultured lymphocytes from HTLV-I carriers, seronegative normal individuals and HAM-derived cells. Some of these cells also showed surface labeling with WT-31. These results indicate that expression of HLA-D and CD25 but not pan-T cell antigens such as CD3 and CD5 on the viral envelope is necessary for lymphocyte transformation and that the viral envelope is negative for alpha/beta chain expression of T-cell receptors.
