Abstract: A major product of oxygen damage to DNA is 8-oxoguanine. The mutagenic potential of this adduct was tested in the M13G assay, a site-specific assay for in vivo mutations caused by guanine adducts inserted into the E coli lacZ gene. Site-specific insertion of 8-oxoguanine using the DNA polymerase activity of HIV-1 reverse transcriptase (which allows promiscuous addition of adducted nucleotides to DNA) was followed by transfection into E coli. The mutation frequency of the 8-oxoguanine-containing DNA was 0.11% (54,000 plaques screened), which was 4 times higher than controls. 40 of the 68 mutants were G-C to T-A transversions, whereas none of these mutations was found among the 9 mutants sequenced from controls (among 37,000 screened). Thus, the predominant single base substitutions caused by 8-oxoguanine are G-C to T-A transversions, suggesting that 8-oxoguanine has the potential to contribute to the occurrence of spontaneous G-C to T-A transversions.
