Abstract: There is justifiable optimism for a renewed intensification of efforts to translate modern immunology and molecular technology into tumor immunotherapeutic protocols. Activating the immune system can cause a response against quiescent tumor cells and lymphocytes can retain long-term memory for tumor-specific reactivity. Tumor immunology is reviewed in four chapters: (1) overview of tumor immunology (antigen presentation, T-cell activation, tumor antigens, and gene transfection); (2) current views on immune surveillance (tumor cell immunogenicity, immune surveillance in operation, and tumor surveillance without immunogenicity); (3) immunologic mechanisms of tumor cell killing (monoclonal antibodies, cellular immunity [antigen-restricted and antigen-unrestricted lymphocytes, monocytes/macrophages and neutrophils, and enhancement of effector cell killing], cytokines, and nonspecific immunostimulants); and (4) malignancies associated with immune deficiencies (malignancies in organ transplant recipients [cancers of the skin and lips, non-Hodgkin's lymphoma, Kaposi's sarcoma, and carcinomas of the vulva and perineum], cancers following cyclosporine therapy, cancers in nontransplant patients (pts) given immunosuppressive therapy, second cancers in pts given cytotoxic therapy, primary immunodeficiency states, cancers in AIDS [Kaposi's sarcoma, lymphomas, and other cancers], and possible causes of cancer in immunodeficient pts).
