CHARACTERIZATION OF IMMUNE DYSFUNCTION IN CATS WITH FELINE LEUKEMIA VIRUS-INDUCED IMMUNODEFICIENCY DISEASE

Abstract: A panel of in vitro assays was used to investigate the nature of the immune dysfunction in cats infected with a naturally occurring FeLV isolate (designated FeLV-FAIDS) which induces a fatal immunodeficiency syndrome in 100% of experimentally infected cats. Concanavalin A-induced blastogenesis, xenogeneic mixed lymphocyte reaction assays, stimulation of lymphocytes by soluble protein antigen, and cytotoxic T lymphocyte assays were used to evaluate T cell and antigen-presenting cell function. During the asymptomatic period preceding immunodeficiency disease, I was unable to detect any significant differences in the responses of T cells from groups of FeLV-FAIDS-infected and normal cats. However, early in the course of disease, humoral immune responses in the FeLV-FAIDS-infected cats were dramatically impaired. As early as 7 weeks postinoculation, the ability to mount either an IgM or IgG response to soluble protein antigens was lost. B cell function, as analyzed by lipopolysaccharide-induced blastogenesis, and relative numbers of B cells, as analyzed by immunofluorescence, were not significantly different between the FeLV-FAIDS-infected and normal cats. It, therefore, appeared that T cell-mediated help of cellular immune responses and B cell numbers and function were unaffected in FeLV-FAIDS-infected cats. These cats were, however, unable to produce antibodies in response to antigenic challenge. These results suggest that FeLV-FAIDS infection impairs a subpopulation of T helper cells whose primary function is to provide help for the production of antibody. This impairment of humoral immunity appears to be a major component of the immunodeficiency syndrome in FeLV-FAIDS-infected cats.
