ZIDOVUDINE: CLINICAL EXPERIENCE IN SYMPTOMATIC HIV DISEASE

Abstract: Zidovudine (AZT) is effective in the treatment of patients with advanced HIV disease, and it is likely that intervention with AZT early in the course of symptomatic HIV infection will prevent or delay progression to AIDS or AIDS-related illnesses and thereby change the natural history of HIV infection as currently recognized. Clinical experience with AZT is summarized under the following headings: clinical pharmacology, efficacy (survival, opportunistic infections, neurological disease, autoimmune thrombocytopenia, and other considerations), immunological monitoring, virological monitoring, adverse reactions (myositis, neurological toxicity, hematological toxicity, and hepatotoxicity), drug interactions, and prophylaxis for the prevention of HIV infection.
