Abstract: Simian immunodeficiency virus (SIV) is a member of the same family of closely related lentiviruses that includes HIV-1 and HIV-2. All of the viruses of this family share similarities in nucleotide sequence and a tropism for CD4-bearing lymphocytes. The AIDS-like disease induced by SIV in rhesus monkeys was used as a model to explore the nature of the T-lymphocyte response following infection with viruses of the HIV family. Activated CD8+ lymphocytes were present in increased numbers in the paracortex of lymph nodes of SIV-infected rhesus monkeys with a lymphadenopathy syndrome, and SIV was isolated more readily from CD8+ lymphocyte-depleted peripheral blood lymphocytes (PBLs) of SIV-infected animals than from their unfractionated PBLs. Rather than reflecting CD4+ lymphocyte-enrichment of the CD8+ lymphocyte-depleted cell populations, the CD8+ cells themselves appear to be critical in this regulatory interaction. In fact, CD8+ lymphocytes from SIV-infected rhesus monkeys, but not uninfected rhesus monkeys, blocked SIV replication in vitro in PBL populations. These findings suggest that the CD8+ cell that exerts this effect is either an activated lymphocyte or a memory lymphocyte. A T-lymphocyte population that blocks replication of viruses of the HIV family could contribute to containing the progression of AIDS. (4 Refs)
