Abstract: The strain specificities of affinity-purified human antibodies and animal antisera to a nonglycosylated HIV-SF2 polypeptide representing the gp120 region of the env genes were examined, and efforts were made to identify the target(s) of the HIV-SF2-neutralizing antibodies that do not bind to this antigen. Neutralizing antibodies in HIV antibody-positive human sera were found to bind to env 2-3, a nonglycosylated polypeptide corresponding to the amino acid sequence of gp120, the large external glycoprotein of the HIV-SF2 isolate. However, there were also HIV-SF2-neutralizing antibodies that did not bind this antigen. Other recombinant antigens from the env and gag genes, produced in genetically engineered yeast, did not bind this second category of HIV-neutralizing antibodies. Human antibodies specific for env 2-3 cross-neutralized some, but not all, HIV isolates. Animal antisera raised to env 2-3 also exhibited restricted neutralization of HIV isolates. The majority of the cross-neutralizing activity of human sera is directed against as yet unidentified epitopes in env (eg, conformational epitopes) or other target molecules. Future studies will be directed toward identifying these target regions in order to elicit a humoral immune response that will be effective against all HIV strains. (22 Refs)
