Abstract: Site-directed serology employing synthetic peptides is one means of improving the possibility of determining type-unique antibodies in a screening assay for HIV-1 or HIV-2. An enzyme-linked immunosorbent assay (ELISA) using such peptides has been developed that is specific for HIV-1. The test, designated the E32/E34 ELISA, uses synthetic antigens representing a portion of the transmembrane glycoprotein, gp41. Attempts to develop an ELISA that is specific for HIV-2 are described. A comparison of simian immunodeficiency virus and HIV sequence data for the transmembrane protein regions revealed a region of homology, and two peptides were synthesized that were homologous to E32 and E34. It was found that one of these (AlaIleGluLysTyrLeuGluAspGlnAlaGlnLeuAsnAlaTrpCysAlaPheArgGlnValC- ys) could be used to categorically separate sera of HIV-1-infected patients from those of HIV-2-infected patients. Correction of a small mistake in the original peptide (missing Gly between Trp and Cys) yielded an increase in both sensitivity and specificity of the ELISA. (8 Refs)
