Abstract: Platelet-derived growth factor (PDGF) and its structure, function, and relationship with the transforming gene product of simian sarcoma virus are discussed under the following headings: the subunit composition of PDGF, amino acid sequence of PDGF; structural and functional relationship between p28sis and PDGF; and expression of the c-sis gene in human tumor cells. Using a 32P-labeled c-sis probe, the authors recently identified c-sis transcripts on Northern blots of polyA+ RNA isolated from human osteosarcoma cell line U-2 OS and other sarcoma and glioma lines. It is conceivable that an undue expression of the c-sis gene and synthesis of PDGF leads to an autocrine stimulation of growth of these tumor cells; it is noted in this connection that gliomas and sarcomas are derived from PDGF-responsive tissues. However, expression of c-sis is not a constant feature of gliomas and sarcomas, indicating that the undue expression of c-sis and PDGF production may represent only one of several mechanisms of constitutive mitogenesis in glioma and sarcoma cells.
