A NOVEL HUMAN LYMPHOTROPIC RETROVIRUS (LAV): NEW DATA ON ITS BIOLOGY AND ROLE IN AIDS

Abstract:
The main characteristics of lymphadenopathy associated virus (LAV) are summarized and the relationship of this virus to acquired immune deficiency syndrome (AIDS) is discussed. LAV1 was isolated from lymph node tissue of a French homosexual with lymphadenopathy syndrome (LAS). LAV is similar to human T-cell leukemia virus (HTLV)-I and II in terms of Mg++ dependence of the reverse transcriptase for optimal reaction and tropism for T lymphocytes. However, LAV differs from HTLV I and II by lack of antigenic cross-reactivity of the major core protein with HTLV-I and II p24, different ultrastructure, and a lack of in vitro transforming activity. LAV antibodies have been detected at high frequency by RIPA and/or ELISA in patients (pts) with AIDS or at high-risk for AIDS, including homosexuals, hemophiliacs, Zairians, Haitians, and iv drug users presenting with AIDS or lymphadenopathy syndrome (LAS). No virus isolation could be achieved from healthy blood donors, laboratory workers, or pts with malignant disease. Results of in vitro studies showed a selective tropism for T4 cells correlated with a cytopathic effect on the target cells. Sero-epidemiological studies of Zairian AIDS pts, French and American AIDS pts, high-risk, but healthy, subjects, representatives of the general population in France, and healthy Zairian mothers are consistent with the involvement of LAV-related viruses in AIDS and LAS.
