Abstract: Peripheral blood lymphocytes (PBL) from eight males with acquired immunodeficiency syndrome (AIDS) and prior histories of genital or anal herpes were treated with monoclonal antibody against OKT8 cells. PBL from each of six patients (pts) tested showed little or no responsiveness to phytohemagglutinin (PHA) stimulation, but exogenous T cell growth factor (TCGF) restored some responsiveness in four. AIDS PBL were unresponsive to antigen from herpes simplex virus (HSV) and did not become responsive in the presence of TCGF. After depletion of OKT8 cells by 70-80% using specific monoclonal antibodies, responsiveness to PHA developed in PBL from 4 of 6 pts. None of the OKT8-depleted cultures responded to HSV. Response to HSV occurred, however, in PBL from 3 of 5 pts which underwent OKT8 depletion and TCGF expansion. The authors conclude that the apparent anergy of AIDS lymphocytes can be reversed by in vitro manipulations, and that these cells may retain functional specificity. (26 Refs)
