Abstract:
Adult T-cell leukemia (ATL) is an endemic malignancy of southwest Japan which has been closely associated with a retrovirus, ATLV, in serologic, epidemiologic, virological, and molecular studies. All criteria for a causal relationship have been met. Mycosis fungoides (MF) and the Sezary syndrome (SS) are T-cell lymphomas in which isolation of the human T-cell leukemia virus (HTLV) has been reported, but serologic, virological, and molecular studies have been inconsistent. Adult T-cell leukemia/lymphoma (ATLL) is a malignancy of helper T-cells; it resembles Japanese ATL but occurs in blacks. In a study of fresh leukemic cells from children with acute lymphoid leukemia, inclusion bodies containing virus-like particles (VLP) were observed in 12 of 28 patients, and VLP were also expressed in leukemic cell lines. The author concludes that ATLV is a causative agent of ATL; that ATLV is not associated with MF or SS; that reports associating HTLV with MF or SS probably misdiagnosed these diseases for ATLL; that ATLV and HTLV are related but not identical; and that VLP may represent the expression of viral structural proteins.
