Abstract: The potential etiologic role of virus, cytogenetic abnormalities, and immunodeficiency states in non-Hodgkin's lymphoma (NHL) is discussed. Recent observations suggesting that some NHLs represent complex examples of the breakdown of normal immune responses are emphasized. These include the occurrence of chromosome 14 abnormalities in B-cell tumors, the association between NHLs and genetically determined immunodeficiency diseases, the role of Epstein-Barr virus in Duncan's familial immunodeficiency disorder, the occurrence of a fatal polyclonal B-cell immunoblastic lymphoproliferative disorder after thymic transplantation in patients with combined immunodeficiency disease, the development of histiocytic lymphomas or immunoblastic sarcomas in renal transplant patients, and the increased incidence of NHLs in patients with autoimmune disease such as Sjogren's disease or systemic lupus erythematosus. Chronic antigenic stimulation leading to unchecked lymphoma proliferation could be an etiologic factor common to hereditary and acquired immunodeficiency diseases, oncogenic viral infections, and autoimmune diseases.
