This patented technology is currently being used in a Phase I clinical trial evaluating a colon cancer vaccine. Essentially, the colon cancer vaccine is comprised of irradiated fibroblasts from a skin biopsy, genetically modified to produce interleukin-2 ("IL-2"), along with irradiated tumor cells excised from the patient.
IL-2, a cytokine which belongs to a class of molecules that regulate the immune and inflammatory responses in humans, can increase the proliferation of T cells. T cells are known to play a role in tumor rejection. In May 1995, the results of preclinical studies using this technology were published in Human Gene Therapy. Seventy-five percent of treated animals developed systemic anti-tumor immunity and were protected against subsequent tumor cell challenges. This study also identified the important relationships between IL-2 dose, immune system effector mechanisms and antitumor efficacy.
"Preclinical studies indicated that this technology may be able to destroy established tumors and prevent recurrence. We are now nearing the completion of our Phase I clinical trial in colon cancer and expect to begin additional clinical trials in 1997 for both colon and prostate cancers using this technology," said Dennis J.
